I am interpreting your question as a need to replace the faucet shut off on the hot side of the water supply in your bathroom and not the faucet. If this is the case you can shut off the incoming cold water supply on the water heater while you replace that shutoff.You will have some hot water in the line but that is normal. I would replace both the cold and hot shutoffs using a newer 1/4 turn type. You should also check the supply lines and insure they are in good condition. This will take about an hour or possibly 1.5 hours but no more than that. I would purchase what is needed first and insure the supply lines are the correct length and are the proper size for your connections.

If you want to turn it iff to change the valve don’t fool with the heater. You must have a master shut off valve that controls the water supply to the whole house. It would be where the water line comes into the house. then drain the taps in the bathroom to remove water from the line and change your valve
yes, if you shut off the water going into the heater, there will be no pressure to push water out. You should be ok without turning off the burner for a short time.
